Key Points

  • The aim is to improve detection methods for the viable pathogen Diaporthe destruens in sweet potato tubers.
  • Utilized BIO-PCR to identify viable Diaporthe destruens from sweet potato tubers.
  • Compared sensitivity of BIO-PCR to traditional real-time PCR methods.
  • Analyzed detection rates of viable and nonviable pathogens.
  • BIO-PCR showed increased sensitivity for detecting viable D. destruens.
  • Traditional PCR did not differentiate between viable and nonviable pathogens.
  • BIO-PCR helps avoid false positive results in pathogen detection.

Abstract

Real-time PCR is highly specific and sensitive for Diaporthe destruens, which causes foot rot of sweet potato. However, PCR detects pathogens whether they are viable or not. We utilized BIO-PCR to detect viable D. destruens from sweet potato tubers. BIO-PCR increased detection sensitivity of the viable pathogen but not the nonviable. Thus, BIO-PCR could be useful to avoid false positive results, which arise from detection of nonviable pathogen.
