Abstract

目的 在水环境小平台法构建的脾虚证模型基础上,利用血清代谢组学探讨脾虚与肿瘤的发病关系。 方法 采用随机数字表法将40只小鼠分为空白组和模型组。造模2周后,通过监测小鼠的一般体征、代谢及行为学变化评估脾虚模型的建立情况。从两组各随机选取10只小鼠进行Lewis肺癌细胞(LLC)皮下接种,未接种者分为正常组和脾虚组,接种者分为肿瘤组和脾虚肿瘤组,记录肿瘤生长曲线。检测各组小鼠脏器指数,对脾脏及肿瘤组织进行HE染色。采集血清样本,经液相色谱-质谱联用(LC-MS)分析,筛选潜在生物标志物并分析相关代谢通路。 结果 与正常组比较,脾虚组小鼠体重下降,抓力与旷场实验指标降低,尿D-木糖排泄率下降,粪便含水率增加,脾脏及胸腺指数减低。HE染色显示,脾虚与肿瘤均显著影响脾脏及肿瘤组织的病理结构,诱发免疫应答,脾虚状态加剧了肿瘤的病理表现。代谢组学分析共筛选出8种与脾虚相关的差异代谢物,涉及甘油磷脂代谢、花生四烯酸代谢等通路;另有8种潜在生物标志物与脾虚对肿瘤的影响相关,涉及甘油磷脂代谢和核苷酸代谢。 结论 脾虚状态可加重肿瘤小鼠的病理损伤,其机制可能与甘油磷脂代谢异常密切相关。
