Novel hPSC protocol generates heart-macrophage assembloids for scalable modeling of immune-cardiac interactions and atrial arrhythmias.
This protocol provides a reproducible and scalable platform for generating human heart-macrophage assembloids to study immune-cardiac interactions and model atrial arrhythmias.
May inform immune-targeted AF therapies; extends scalable modeling of immune-cardiac interactions.
Here, we present a protocol to generate fully human pluripotent stem cell (hPSC)-derived human heart-macrophage assembloids. We describe steps for differentiating hPSCs into monocytes, generating cardiac organoids, integrating CD14 + monocytes to become cardiac tissue-resident macrophages, and maturing the assembloids with metabolic cues. Furthermore, we provide a use-case example of disease modeling for the study of atrial arrhythmias. The system provides a reproducible, scalable, and efficient platform for studying immune-cardiac interactions, inflammation, and arrhythmogenesis. For complete details on the use and execution of this protocol, please refer to O’Hern et al. 1 • Steps to generate monocytes from hPSCs using a scalable EB-based differentiation workflow • Instructions for producing human heart organoids from matched hPSC lines • Guidance on assembling monocytes with heart organoids to form hHMAs • Steps to induce atrial arrhythmias in hHMAs using chronic inflammatory stimulation Publisher’s note: Undertaking any experimental protocol requires adherence to local institutional guidelines for laboratory safety and ethics.
