A one-pot synthesis improves α-amino ketones in pharmaceutical compounds, suggesting efficiency in drug development.
An efficient one‐pot strategy has been developed to synthesize α‐amino ketones from ketones and amines using dimethyl sulfoxide and oxalyl bromide. The mechanistic studies indicated that α‐bromination of ketones is proposed as the key step, followed by nucleophilic substitution of with amines. The practicality of this strategy is highlighted through one‐pot gram‐scale synthesis of two vital pharmaceutical compounds, pyrovalerone, and amfepramone.
