Findings reveal STK24-expressing cells influence tumor progression, implying potential for enhancing immunotherapy responses.
in vitro/in vivo) tracked via fluorescence imaging/FCM.Tumor tissues underwent scRNA-seq and bioinformatic analysis.Human MPE samples from lung cancer patients yielded autologous MW-TMPs, tested in 3D spheroids and zebrafish PDX models. Results: This method yields more MW-TMPs with lower nucleic acid content than UV-TMPs.MW-TMPs are HMGB1-enriched and potently induce ICD in tumor cells. In lung adenocarcinoma models, MW-TMPs showed superior antitumor activity by reprogramming the immunosuppressive microenvironment: activating dendritic cells, promoting M1 macrophage polarization, and expanding cytotoxic NK/T cells.MW-TMPs delivered methotrexate to tumors and synergized with anti-PD-L1 therapy.Efficacy was validated in patient-derived models (MPE 3D spheroids, zebrafish xenografts). Conclusions:Microwave irradiation provides an efficient, safe, scalable method for therapeutic TMP production.MW-TMPs function dually as a potent immunotherapeutic (inducing ICD, activating innate/adaptive immunity) and targeted drug delivery nanoplatform.Their synergy with immune checkpoint blockade highlights strong translational potential for clinical cancer therapy.
