Key Points

  • This research aims to evaluate the efficacy, safety, and pharmacokinetics of semaglutide 1.7 mg in treating obesity in adolescents.
  • Conducted PK/PD and safety modelling for semaglutide 1.7 mg
  • Compared results in adolescents versus adults and 2.4 mg dosage
  • Analyzed tolerability and BMI reduction
  • 1.7 mg semaglutide shows comparable exposure and tolerability in adolescents
  • Meaningful reduction in BMI in adolescents
  • Supports inclusion in Wegovy label without needing a dedicated placebo trial

Abstract

PK/PD and safety modelling indicated 1.7 mg semaglutide provides comparable exposure, tolerability and meaningful BMI reduction in adolescents versus adults and versus 2.4 mg. These results supported approval of 1.7 mg for adolescents ≥ 12 years in the November 2024 Wegovy label, obviating a dedicated adolescent 1.7-mg placebo trial.
