In the Research Article (e26126), Ling-Yi Kong, Kai-Bo Wang, and co-workers reveal that 8-oxoguanine, the most prevalent oxidative DNA lesion, induces precise, context-dependent structural remodeling of G-quadruplexes (G4s) in human gene promoters. They proposed that G4s act as sensors of oxidative damage and contribute to cellular responses to oxidative stress. This mechanistic insight opens new therapeutic avenues for oxidative stress-associated diseases, including cancer and neurodegenerative disorders.
