This research reports a Pd-catalyzed cascade cyclization for constructing complex fused scaffolds, highlighting new synthetic pathways.
Harnessing metal-catalyzed cascade cyclizations provides powerful strategies for accessing complex polycyclic scaffolds from linear feedstocks. We present herein a Pd(II)-catalyzed cascade cyclization of 1,8-enyne oxime ethers via a consecutive intramolecular oxy-palladation/Heck-type coupling for isoxazole-oxocine based di-nuclear scaffolds. The presence of BINAP-ligand diverges the pathway to the exo-mode, which followed a C-H annulation for the construction of oxepane fused trinuclear polycyclic scaffolds. Some control experiments were carried out to understand the mechanism.
