Fenugreek seed extract attenuates doxorubicin-induced cardiorenal and hepatic toxicities by boosting antioxidant activity.
Why the study?
The biochemical, antioxidant, and histological efficacy of fenugreek seed extract against doxorubicin-induced hepatotoxicity, nephrotoxicity, and cardiotoxicity in rats was investigated.
Animal study demonstrates fenugreek seed extract protects against multiorgan toxicity in rats, highlighting its therapeutic potential as an antioxidant adjuvant.
Abstract:The main aim of this investigation is to investigate biochemical, antioxidant, and histological efficacy of fenugreek seed extract against doxorubicin cause hepatotoxicity, nephrotoxicity and cardiotoxicity in rats. Sixty rats were selected and divided into 6 groups (n= 10 per group)Group I: administrated 1 mL isotonic saline orally once daily for 30 days. Group II: received fenugreek seed extract orally 200 mg/ kg b. wt once daily for 30 days. Group III: administrated fenugreek seed extract orally 400 mg / kg b. wt once daily for 30 days. Group IV: intoxicated doxorubicin 4 mg/kg b .wt I/P injection once weekly for 30 days. Group V received fenugreek seed extract 200 mg /kg b. wt followed by doxorubicin I/P once weekly for 30 days. Group vI: received 400 mg /kg b. wt fenugreek seed extract followed by intoxication with doxorubicin once weekly.Serum biochemical analysis revealed Doxorubicin –intoxicated rats showed increases in liver biomarker( ALT, AST, ALP, TC and TG) , decline in TP, Alb, and HDL, along with elevated kidney biomarker(urea, creatinine and uric acid) and heart biomarker (CK-MB and troponin) compared with control group.. Oxidative parameters results exhibit elevation in MDA levels and lowering of SOD and CAT activities in doxorubicin-intoxicated rats. But,Fenugreek protective group lowered MDA and enhanced SOD and CAT activities. Histopathological examination confirmed the biochemical findings, demonstrating preservation of liver, kidney, and heart architecture in fenugreek-protective rats. we concluded that fenugreek exhibits potent antioxidant activity and provides effective protection against doxorubicin-induced hepatic, renal, and cardiac toxicities

rhman et al. (2025) studied Doxorubicin-induced hepatic, renal, and cardiac damage (n=60). Fenugreek seed extract vs. Doxorubicin alone or isotonic saline was evaluated on Biochemical, antioxidant, and histological markers of toxicity. Fenugreek seed extract provided effective protection against doxorubicin-induced hepatic, renal, and cardiac toxicities by lowering MDA and enhancing SOD and CAT activities.
