Novel affinity labels for human O6-alkylguanine-DNA alkyltransferase (hAGT), leading to specific biotinylation of this DNA repair protein (see schematic picture), have been synthesized and characterized. These chemical probes can be used for the specific detection of active hAGT. In addition, a system based on these probes and the display of active hAGT on phage λ has been introduced, which can be applied for the directed molecular evolution of hAGT by using in vitro selection.
