CD71, expressed on the blood-brain barrier (BBB), facilitates receptor-mediated transcytosis—a natural pathway for brain entry. Targeting CD71 offers potential for CNS drug delivery. The SmocMab platform generates fully human monoclonal antibodies by replacing mouse endogenous IgH and Igκ loci with compact human counterparts. To develop fully human anti-CD71 monoclonal antibodies with BBB-penetrating capability, both wild-type C57 and SmocMab mice were immunized with recombinant human CD71 extracellular domain.
