• Zeolitic imidazolate frameworks (ZIFs) have the advantages of excellent stability and decent biocompatibility . • Zeolitic imidazolate frameworks (ZIFs) have promising applications in drug delivery and disease therapy. • This review described the latest advances in the application of ZIF-8 nanoparticles in drug delivery. Diseases have a wide and far-reaching impact on human society, threatening not only individual health but also having a significant impact on the economy, social structure and psychological state. However, in the face of the multiple challenges posed by disease, traditional treatments often struggle to achieve the desired results, and in recent years the emergence of nanotechnology has provided new solutions for the treatment of disease. As a member of the metal–organic frameworks (MOFs) family, the continuous development of zeolitic imidazolate frameworks (ZIFs) and their unique properties, such as tunable pore size, high specific surface area, excellent stability, biodegradability, and good biocompatibility, have led to their promising applications in biomedicine, especially in drug delivery and disease treatment applications. The high porosity of ZIF-8 offers the opportunity to encapsulate large quantities of therapeutic drugs, enzymes, proteins, fluorochromes, or combinations thereof to build novel multifunctional drug delivery systems (DDS). Although similar topics have been reviewed previously, regular updates on this popular topic are particularly necessary due to the rapid development and dynamic changes in the field. This review summarises new and innovative strategies for the construction of ZIF-8-based smart delivery systems in recent years and describes the advances in the application of ZIF-8 in the treatment of diseases. Finally, the future research direction and application prospect of ZIF-8-based nanomedicines are outlined.
