Peripheral UPS with MDM2 amplification matches conventional DDLPS survival, suggesting they are actually dedifferentiated liposarcomas.
Why the study?
Are peripheral undifferentiated pleomorphic sarcomas with MDM2 amplification actually dedifferentiated liposarcomas?
Cohort (n=234)
Yes
Are peripheral undifferentiated pleomorphic sarcomas with MDM2 amplification actually dedifferentiated liposarcomas?
Absolute Event Rate: 93.3% vs 90.7%
Peripheral UPS with MDM2 amplification share clinical, morphological, genomic, and survival characteristics with peripheral conventional DDLPS, suggesting they are in fact DDLPS and should be evaluated with systematic IHC for MDM2.

Peripheral UPS with MDM2 amplification may represent DDLPS; supports reclassification but remains hypothesis-generating without practice change.
Guellec et al. (2014) conducted a cohort in Peripheral undifferentiated pleomorphic sarcomas and dedifferentiated liposarcomas (n=234). Peripheral UPS with MDM2 amplification vs. Peripheral conventional DDLPS and UPS without MDM2 expression was evaluated on 2-year overall survival. Peripheral UPS with MDM2 amplification demonstrated a 2-year overall survival of 93.3% compared to 90.7% for conventional DDLPS, strongly suggesting they are in fact dedifferentiated liposarcomas.
