Measurement of serum prolactin in rats using radioimmunoassay and Nb2 lymphoma cell bioassay revealed different pulsatile patterns, suggesting different forms of prolactin in the circulation.
Different forms of rat prolactin are present in the circulation, as evidenced by different pulsatile patterns detected by radioimmunoassay versus bioassay.
Assay discrepancies may affect prolactin interpretation; leaves open isoform relevance to human physiology.
Serum prolactin (rPRL) was measured by radioimmunoassay (RIA) and the Nb2 lymphoma cell bioassay (Nb2BA) in serial samples collected from rats with chronically implanted intraatrial catheters. In all animals the episodic nature of rPRL secretion was evident in the results obtained using either assay. However, the pulsatile patterns defined by the two assays were different in shape, amplitude and timing of secretory spikes. These results provide compelling evidence that different forms of rPRL are present in the circulation.

Klindt et al. (1982) studied this question. Radioimmunoassay (RIA) and Nb2 lymphoma cell bioassay (Nb2BA) was evaluated on Episodic secretory patterns of rat prolactin (shape, amplitude, and timing of secretory spikes). Measurement of serum prolactin in rats using radioimmunoassay and Nb2 lymphoma cell bioassay revealed different pulsatile patterns, suggesting different forms of prolactin in the circulation.
