1715-P: Health-Related Quality of Life (HRQOL) in People with Type 2 Diabetes (T2D) Receiving the Once-Daily, Oral, Nonpeptide GLP-1RA Orforglipron in ACHIEVE-2 and -3

Introduction and Objective: Significant improvements in HbA1c and body weight were seen with orforglipron (OFG) vs dapagliflozin (DAPA) and oral semaglutide (SEMA) in adults with T2D in ACHIEVE-2 and -3 (A-2 and A-3), respectively. The HRQoL of people treated with OFG vs these oral therapies was measured in these Phase 3, randomized, open-label trials. Methods: Adults with T2D inadequately controlled with metformin were randomized to OFG 3, 12, or 36 mg or DAPA 10 mg QD for 40 weeks (A-2) or to OFG 12 or 36 mg or SEMA 7 or 14 mg QD for 52 weeks (A-3). HRQoL was measured at baseline and Week 40 (A-2) or Week 52 (A-3). Selected PROs including IWQOL-Lite-CT, APPADL, IW-SP, and EQ VAS are reported. The Simplicity of Diabetes Treatment (SIM-Q) was measured at Weeks 4 and 24 in A-3. Results: Improvements from baseline were seen in all HRQoL measures in all treatment groups at Weeks 40 (A-2) and 52 (A-3) (all p<0.05; Table). In A-3, improvements were greater in IWQOL-Lite-CT total score with OFG 12 mg vs SEMA 7 mg, and in EQ VAS score with OFG 36 mg vs SEMA 7 and 14 mg (all p<0.05). No other statistically significant between-group differences were seen in either study. 96% of adults receiving OFG reported it was a simple or very simple treatment for T2D in A-3 at 24 weeks. Conclusion: Oral treatment with OFG, DAPA, or SEMA for 40 or 52 weeks resulted in improved HRQoL from baseline for adults with T2D, with limited between-group differences. Disclosure K.
