Examines immune response shifts in melanoma tumors treated with engineered probiotics, suggesting new immunotherapy avenues.
This dataset comprises single-cell RNA sequencing profiles of B16-F10 melanoma tumors harvested from immunocompetent C57BL/6 mice following intravenous administration of either control EcN (Escherichia coli Nissle 1917) or EcN-SpNA, an engineered probiotic strain secreting the catalytic domain of Streptococcus pneumoniae sialidase A (SpNA). Two biological replicates per treatment group were analyzed. Libraries were constructed using the DNBelab C-TaiM 4 single-cell droplet system and sequenced on an MGI DNBSEQ-T7 platform (paired-end, 150 bp). Raw FASTQ files and filtered gene-cell count matrices (mm10 reference) are provided. This dataset enables investigation of SpNA-mediated desialylation-induced remodeling of the tumor immune microenvironment, including shifts in myeloid and lymphoid cell composition, activation of inflammatory pathways, rewiring of ligand-receptor interactions, and neutrophil subset heterogeneity. The data support the finding that targeted bacterial delivery of sialidase converts immunologically "cold" tumors into "hot" inflamed tumors.
