Shows that spreading depolarizations primarily begin in the somatosensory cortex during energy deprivation, suggesting metabolic vulnerabilities.
Spreading depolarizations (SDs) contribute to acute brain lesion development. While SDs arise in metabolically compromised tissue, regional determinants of SD-susceptibility remain incompletely understood. Here, we show that SDs preferentially originate in the primary somatosensory cortex (S1) during oxygen-glucose deprivation. Notably, before SD initiation, S1 exhibited a more pronounced decline in ATP compared with other brain regions, suggesting S1-specific metabolic vulnerability as a novel working hypothesis underlying SD initiation.
