Supplementary File 2: Limitations and Proposed Solutions ABSTRACT Background: Oncological recurrence is driven by dormant Cancer Stem Cells (dCSCs) hibernating within the bone marrow endothelium. These G0/G1 quiescent cells are resistant to conventional cytotoxic therapies due to upregulation of TGF-Beta, BMP and Wnt signaling cascades. Methods: We propose a novel, non-toxic, multi-modal framework: Phytochemical-Sensitized Resonance Therapy (PSRT). The model integrates: 1) Autologous exosomes co-encapsulating Nigella sativa-derived Thymoquinone (TQ) and SPIONs, 2) CXCR4-mediated biological homing combined with external magnetic guidance, 3) LIFU-triggered payload release, and 4) Cyclotron-resonant PEMF tuned to the bioelectric signature of dCSCs. Results: Volumetric analysis confirms 98.31% of a 100 nm exosome remains available for TQ payload after loading 5x15 nm SPIONs. Nernst calculations identify Kir3.4 / KCNJ9 channels and a -90.84 mV resting membrane potential as the electrophysiological signature of dCSCs. Application of 35 Hz PEMF, derived from Liboff’s cyclotron resonance equation, is predicted to induce selective depolarization and mitochondrial-mediated apoptosis. Conclusion: PSRT provides a theoretical, testable roadmap to eradicate dCSCs without systemic toxicity, offering a potential electroceutical and nano-botanical strategy to prevent cancer relapse.
