Novel strain GXU_MW_L88 T produces vitamin B12 in coral Porites sp., highlighting coral microbiome diversity.
We isolated a novel strain GXU_MW_L88 T from coral Porites sp. Strain GXU_MW_L88 T was Gram-stain-negative, aerobic, non-spore-forming, short rod, with a capacity to produce vitamin B 12 . The growth requirements of this strain were a temperature range of 20–37 °C, a pH range of 5.0–11.0 and a NaCl concentration of 0–6% (w/v). Phylogenetic analysis based on 16S rRNA gene sequence showed that GXU_MW_L88 T shared an identity of 92.2% with Lutimaribacter marinistellae SF-12 T . The highest maximum average nucleotide identity was 72.13% to Paracoccus sediminicola SCSIO 76264 T , while the highest average amino acid identity was 62% to ‘ Oceanomicrobium pacificus ’ KN286. Analyses of both 16S rRNA gene and genome sequences indicated that GXU_MW_L88 T formed a unique branch within the Rhodobacterales . The genomic DNA G+C content of strain GXU_MW_L88 T is 61.3%. The Q-10 was identified as the sole respiratory quinone, while C 16 : 1 ω 7 c and iso-C 10 : 0 3-OH constituted the predominant fatty acids (≥10%). The polar lipids were phosphatidylglycerol, an unidentified amino lipid, an unidentified aminophospholipid, two unidentified phospholipids and six unidentified lipids. Strain GXU_MW_L88 T can produce l -hydroxocobalamin with a yield of up to 284 µg ml −1 . Based on the above features, strain GXU_MW_L88 T is suggested to represent a novel species within a novel genus under the order Rhodobacterales , for which the name Jujubicoccus corallicola gen. nov., sp. nov. is proposed, with the type strain GXU_MW_L88 T (=KCTC 8863 T =MCCC 1K09341 T ). The new family Agaricicolaceae fam. nov. is also proposed phylogenomically to accommodate the genera Agaricicola and ‘ Polymorphum ’.
