Antimicrobial resistance (AMR) is a pressing global health challenge, threatening individuals across all ages and regions. Antibiotics, crucial for reducing infant mortality and facilitating medical treatments such as surgeries and chemotherapy, are increasingly ineffective due to the rise of multidrug-resistant bacteria. According to the WHO, antibiotic resistance is among the top ten threats to global health, exacerbated by factors such as overuse, misuse, and agricultural application of antibiotics. The mortality rate attributable to AMR exceeds 1.2 million annually, surpassing deaths caused by HIV and malaria. Developing countries bear the greatest part of this burden, although developed nations are not spared either. The primary pathogens contributing to AMR include Pseudomonas aeruginosa , Acinetobacter , Staphylococcus aureus , Klebsiella pneumoniae , and Escherichia coli . Resistance to first-line antibiotics, including beta-lactams and fluoroquinolones, accounts for over 70% of deaths associated with bacterial infections. Drivers of AMR include overuse of antibiotics often due to unregulated access and inappropriate prescribing misuse, leading to genetic mutations and increased virulence; and extensive use in agriculture, where resistant bacteria can enter the human food chain. Compounding the problem is the stagnation in the development of new antibiotics. Gram-positive bacteria, such as Staphylococcus aureus and Enterococcus species, and Gram-negative pathogens like Klebsiella pneumoniae and Escherichia coli, represent significant threats. These bacteria use various mechanisms to develop resistance that include efflux pump, production of degrading enzymes like beta-lactamase production and finally alteration of metabolic pathways. The development of multidrug-resistant strains underscores the urgency of a global response. Effective measures include regulating antibiotic use, improving sanitation, and fostering innovation in drug development to combat this growing crisis.
