One-node potato (Solanum tuberosum L. cv. ‘Katahdin’) cuttings were used to study early anatomical changes associated with tuberization. Starch deposition and the percentage frequency of cells in mitosis increased in the medullary region of the bud within 1 d after cutting, whereas increases in average cell size were not detected until 4 d after cutting. Starch deposition and mitosis were the earliest detectable changes in anatomy associated with tuber initiation.
