The selective modification of biomolecules has been transformed from a distant challenge into a central pursuit in chemical biology. Over the years, click reactions, transition metal catalysis, and visible-light photocatalysis have each expanded the chemist’s ability to engage with nature’s most intricate molecules. Metallaphotoredox catalysis now bridges the gap between these approaches, merging the reactivity of transition metals with the precision and mildness of light-driven processes. This synergy allows site-selective transformations of proteins, nucleic acids, and carbohydrates that were once considered out of reach. In this Perspective, we examine how metallaphotoredox catalysis has reshaped the bioconjugation toolbox by highlighting recent breakthroughs and discussing opportunities, remaining challenges, and future directions.
