A device which features the intermarriage of HPLC components with those originally designed for autoanlysis or amino acid analysis is described. Using this device polyamines in tissue extracts have been separated in 65 min by cation exchange chromatography using a stepped sodium gradient. Postcolumn fluorescence derivatisation with o-phthalaldehyde gives a linear range from 100 pmol-90 nmol with a coefficient of variation of 2% at 9 nmol. Technical problems encountered and important factors influencing performance are fully described.
