The advantages of analgesia using combinations of drugs are discussed with reference to the pharmacological interactions of opioid agonists with three other classes of drugs: α2-adrenoceptor agonists, local anaesthetics and N-methyl-D-aspartate receptor antagonists. The principles behind the determination of synergy are also discussed.
