The National Communicable Disease Center micro antistreptolysin O test and the conventional Rantz-Randall antistreptolysin O tube test were performed with 396 human serum samples to determine the reproducibility of each test and correlation of titers between the two tests. Reproducibility of each test was good, with less than 2% of the titers in duplicate sera differing by more than one dilution step. The difference between the dilution schemes of the two tests makes it difficult to compare the titers; however, the general correlation between the titers was good. A comparison of the geometric mean titers gave a linear correlation coefficient of 0.94. Differences in titer by the two methods appear to be due primarily to “scale” differences in the two dilution schemes.
