Testosterone (T) levels rose in 7 of 9 adult males given iv doses of synthetic LRF ranging between 0.1–450 μg. 10 μg was the smallest dose to stimulate a consistent rise of LH and a statistically significant T elevation. The magnitude of the net T rise varied between 740–3979 pg/ml and was always greater than spontaneous fluctuations measured during a 3-hr control study in each subject. The larger LRF doses tended to elicit the greater responses; however, no dose response relationship could be established. The time lag between the induced LH peaks and maximal T rises varied between 30–100 min (median 90 min). LRF injections were followed by a small rise in androstenedione levels in 4 of 9 subjects. Similarly, a dose-response relationship was not found.
