A method is presented for the isolation of mutants of the obligate thermophile Bacillus stearothermophilus. Mutants blocked in fumarase, aconitase, and alcohol dehydrogenase have been isolated by the procedures described. The availability of thermophilic mutants provides a genetic approach for studying the phenomenon of thermophily.
