Investigates BrMYB51 gene localization and its effects on glucosinolate levels in plants, indicating significance for crop improvement.
The MYB51 transcription factor plays a pivotal role in regulating the biosynthesis of indolic glucosinolates in plants. This study investigates BrMYB51 , focusing on its sequence information, expression patterns, and subcellular localisation of the encoded proteins. Using genomic sequence data from Brassica rapa , we successfully amplified the full‐length coding sequence (CDS) of the BrMYB51 gene. Analysis of the BrMYB51 promoter revealed an abundance of cis‐elements associated with plant hormone responses. Expression profiling demonstrated that BrMYB51 was induced by abscisic acid (ABA), methyl jasmonate (MeJA), and salicylic acid (SA). Phylogenetic analysis indicated that BrMYB51 shares the closest genetic relationship with MYB51 proteins from Brassica napus and Raphanus sativus L. Subcellular localisation experiments using Agrobacterium tumefaciens ‐mediated transient expression showed that the green fluorescent protein (GFP) – fused BrMYB51 was localised to both the nucleus and plasma membrane in the Nicotiana benthamiana leaves. Overexpression of BrMYB51 in transgenic Arabidopsis resulted in elevated levels of indolic glucosinolates, suggesting that BrMYB51 enhances glucosinolate content by regulating the expression of genes involved in the indolic glucosinolate biosynthesis pathway.
