Monolayer cell cultures of calcitoninsecretingcells have been derived from trout ultimobranchialglands. These cultures have been used to study calcitonin secretion and its regulation in vitro; calcitonin has been measured by radioimmunoassay. The cultures exhibit linear hormonalsecretion rates and secretory responses to several substances at concentrations as low as 10–8M. Pancreozymin, prostaglandins and dibutyryl cAMP cause at least a 5-fold increase in calcitonin secretion;calcium has a small secretory effect. This system affords a unique method for the study of both the extracellular and intracellular factors involvedin the regulation of calcitonin secretion. (Endocrinology95: 1142, 1974)
