Demonstrates enhanced treatment efficiency in pancreatic ductal adenocarcinoma, indicating a novel therapeutic approach.
Optimized Bicomposite Nanotherapeutic Architecture for Pancreatic Ductal Adenocarcinoma (PDAC): A High-Performance In Silico Framework (v5.0) Background: Pancreatic Ductal Adenocarcinoma (PDAC) remains one of the most lethal malignancies, largely refractory to conventional therapies due to an exceptionally dense desmoplastic stroma and robust cellular resistance mechanisms. To overcome these formidable physiological barriers, this project presents an advanced nanotherapeutic framework utilizing a bimodal bicomposite architecture optimized to a 60 nm hydrodynamic diameter (rH = 30 nm). Methodology: The platform deploys a coordinated, two-wave administration strategy. Wave I utilizes hypersuccinylated nanoparticles (NP-A, ζ = -40 mV) engineered to achieve electrostatic "Brownian slip" and stromal fluidization, simultaneously delivering circ-shRNA to silence Glutathione Peroxidase 4 (GPX4) and prime the tumor microenvironment (TME) via massive lipid hydroperoxide accumulation. Wave II deploys a logic-gated retention vector (NP-B, 80 nm) equipped with a multi-layer molecular biocomputing circuit. This circuit integrates structural [AND] gates (dependent on extracellular MMP-2/MMP-9 and intracellular Cathepsin B) to release the pan-caspase inhibitor Z-VAD-FMK, alongside [NOT] gates driven by miR-122 recognition sequences to guarantee $>95%$ off-target exclusion in healthy hepatic tissue. High-Performance Computational Core (Project60 v5): To bridge theoretical design with empirical validation, version 5.0 integrates a hardware-accelerated numerical solver powered by JAX (@jax.jit). This computational core models 3D finite-difference mass transport, extracellular matrix dynamics, and ferroptotic kinetic grids. Furthermore, native parsing of single-cell and spatial transcriptomics (.h5ad / AnnData format) maps actual topological features into an interactive Streamlit/Plotly web dashboard (Project60 TME Explorer), enabling real-time simulation of therapeutic accumulation and cellular viability across a 12 to 168-hour temporal horizon. Safety & Biosecurity: Systemic biosafety is reinforced by an orthogonal, antibiotic-free mechanism: a Theophylline-responsive synthetic riboswitch capable of achieving a $99%$ cessation of protein elongation within two hours of ligand administration, acting as an instantaneous clinical emergency brake. Conclusion: The CAMPA v5.0 framework establishes a rigorously validated, high-performance in silico foundation for translational oncology (TRL 3-4). By redirecting tumor cell fate away from apoptosis toward immunogenic, non-caspase-dependent ferroptosis—thereby neutralizing the mitogenic "Phoenix Rising" repopulation effect—the architecture offers a precise, safe, and computationally reproducible paradigm for eradicating chemoresistant solid tumors.
