3212 Introduction: <w:sdt id="953907347" sdttag="goog_rdk_1">High resolution scans of immunohistochemical (IHC) stains of Alzheimer’s disease (AD) brain slices and autoradiography binding assays both give us information about the distribution of Tau protein and ꞵ-Amyloid. Accurate assessment of the amount and regional location of Tau and ꞵ-Amyloid and is essential to understand AD pathology. Our goal is to quantitatively assess IHC-autoradiography images with their respective autoradiographic binding assay to investigate the relationship between Tau and ꞵ-Amyloid distributions with [</w:sdt>¹²⁵I]IPPI and [¹⁸F]Flotaza binding, respectively. Methods: Postmortem human cortex from AD subjects were IHC stained for total Tau, ꞵ-Amyloid, and [¹²⁵I]IPPI, [¹⁸F]Flotaza autoradiography for Tau and ꞵ-Amyloid as reported. Using Qupath, a pixel thresholder was created to outline the IHC images. Using Tau and ꞵ-Amyloid annotations for training, pixel classifiers were generated to measure area of Tau and ꞵ-Amyloid in each slice, respectively. The Tau and ꞵ-Amyloid detection objects were automatically created, and objects outside the identified bounds for area and DAB intensity were deleted using modified scripts to increase the accuracy of object detection. Binding values from the autoradiographic studies in the form of gray matter DLU, area of Tau and ꞵ-Amyloid, number of Tau and ꞵ-Amyloid detections were analyzed and plotted in MATLAB. Results: Figure 1A shows the area of Tau plotted against the autoradiography [¹²⁵<w:sdt id="1057353433" sdttag="goog_rdk_6">I]IPPI binding. Tau area and </w:sdt>[¹²⁵<w:sdt id="1057353433" sdttag="goog_rdk_6">I]IPPI binding had a correlation coefficient of .7132. </w:sdt><w:sdt sdttag="goog_rdk_6">Similarly, ꞵ-Amyloid area was plotted against the [¹⁸F]Flotaza binding in Figure 1B, with the correlation coefficient being .7437. Figure 1C shows the number of Tau detections in gray matter plotted against the IPPI binding DLU/mm^2 in gray matter, with a correlation coefficient of .8285. Figure 1D shows ꞵ-Amyloid detections plotted against [¹⁸F]Flotaza binding DLU/mm^2 in gray matter which yielded a correlation coefficient of -.4994. </w:sdt> Conclusions: Compared to semi-quantitative histopathological scoring, our novel method of analyzing immunohistochemical stains with autoradiography binding assays has the potential for quantitative image analysis of IHC with autoradiography for Tau and ꞵ-Amyloid in AD. Efforts are underway to specify detection of possible isoforms of Tau and ꞵ-Amyloid which may account for size differentiation.
