Tranilast significantly reduced cardiac extracellular matrix deposition and the expression of TGF-β1, collagen type I, and collagen type III in a mouse model of radiation-induced heart fibrosis (P<0.01).

The heart is a major dose-limiting organ for radiotherapy of cancer in the thoracic region. The purpose of this study was to examine the protective effect of tranilast on the radiation-induced heart fibrosis model using the C57BL/6 murine strain. A significant reduction in the expression of TGF-<TEX>β1</TEX>, collagen type I and collagen type III was observed in the radiation plus tranilast group. The authors also suggest the use of tranilast in a clinical trial for the prevention of radiation-induced heart fibrosis.

S.H. Moon (2012) studied Radiation-induced heart fibrosis (n=16). Tranilast vs. Radiation alone was evaluated on Cardiac extracellular matrix deposition and expression of TGF-β1, collagen type I, and collagen type III (p=<0.01). Tranilast significantly reduced cardiac extracellular matrix deposition and the expression of TGF-β1, collagen type I, and collagen type III in a mouse model of radiation-induced heart fibrosis (P<0.01).
