The rate and extent of the oxidation of 17α3H-estradiol was measured in 5 men and 4 women by following the appearance of tritium water in the body water pool. The kinetics of oxidation were multiphasic in character with an initial very rapid stage of 5–10 min common to both sexes. Subsequent oxidation was much slower in men than in women and the total oxidized was 86% of the dose in women vs only 54% in men. The unoxidized material was only minimally excreted in urine which in both men and women contained only about 3% of the administered radioactivity, mostly in the form of 17α3H-estradiol glucosiduronate. The results provide strong evidence of a significant sex difference in the disposition of administered estradiol in man.
