To determine the timing and distribution of bovine viral diarrhea virus (BVDV) antigen in maternal reproductive organs, placenta, and fetal tissues during initial transplacental transmission.
Inoculated seven pregnant Norwegian Red heifers at gestational day 85/86 with noncytopathogenic BVDV and examined tissues following slaughter at days 7, 10, 14, 18, or 22 postinoculation (pi).
Evaluated cryostat sections of maternal uterus, ovaries, placentomes, intercotyledonary fetal membranes, and fetal organs using immunohistochemistry and double immunofluorescence targeting BVDV antigen alongside CD45 and vimentin.
BVDV antigen was first detected in fetuses at 14 days pi, specifically within pulmonary mesenchymal cells and hepatic megakaryocyte-like cells.
Viral antigen appeared in intercotyledonary fetal membranes at 18 days pi and in placentomes at 22 days pi, while remaining undetectable in maternal tissues throughout the study period.
