Feeding intact rats an iodinedeficientdiet for prolonged periods or propylthiouracilfor 10 or more days markedly stimulated in vitro labeled iodothyronine synthesis bytheir thyroid lobes. In vivo administration of 5 UTSH twice daily for 2 days also improved in vitro131I-iodothyronine synthesis in both intactand hypophysectomized rats, but to a lesser extentthan the above dietary regimens. The stimulation of in vitro131I-iodothyronine synthesis isprobably due to a combination of prolongedTSH stimulation of the thyroid gland in vivo combined with iodine depletion of the gland. In vitro triiodothyronine synthesis is apparentlymore dependent on TSH than is that of thyroxine.Earle’s solution seems to be a more satisfactorymedium for iodothyronine synthesis than isKrebs-Ringer phosphate.
