We have analyzed in this study the selectivity of an overoxidized polypyrrole/superoxide dismutase (SOD) coated platinum electrode towards the biosensing of superoxide anion in phosphate buffer solution (pH 7.4). We have dismanteled the different steps involved in the construction of this biosensor to provide a technical examination of its way of working. Our investigation showed that if the overoxidized polypyrrole coating is not permeable to hydrogen peroxide from the bulk solution, no amperometric signal due to the oxidation of hydrogen peroxide formed within the coating by catalyzed disproportionation of O2− by SOD is observed. This may be explained in terms of anion-exclusion behavior of the overoxidized polypyrrole coating.
