A general method has been developed for the analysis of urinary acids by gas chromatography using the methyl ester-trimethylsilyl ether (ME-TMSi) derivatives in a temperature programmed separation with a 6 ft 10% F-60 column. New methods have been developed for the preparation of the trimethylsilyl ester-trimethylsilyl ether derivatives using bis-trimethylsilylacetamide (BSA). When the new silylating agent is used in connection with the new thermostable liquid phases, OV-1 and OV-17, it is possible to program separations to 320°–340°C before the column bleed becomes excessive. The range of compounds that can be studied by gas phase analytical techniques has been extended to include many additional acidic compounds of biological interest such as ether glucuronides, sugar phosphates, and acidic water soluble vitamins.
