Obesity has emerged as a growingly severe public health challenge across the globe. Hemicelluloses, as crucial medicinal components of Panax ginseng, exhibit inhibitory activity against obesity. In the study, the inhibition of α‐glucosidase activity was used as the index of investigation, and a novel extraction method of ginseng hemicelluloses (GSH1) with obesity inhibitory activity was preferred by single‐factor experiments and response surface methodology. Subsequently, the chemical structure of GSH1 was confirmed by GPC, HPLC, FT‐IR, and NMR. Finally, the mechanism of action was explored by circular dichroism spectroscopy, fluorescence spectroscopy, and molecular docking simulations. The antiobesity activity of GSH1 was investigated using in vivo and in vitro obesity models. Results indicated that GSH1 is an acidic polysaccharide with a β‐D‐xylose structure. GSH1 showed significant inhibitory activity to α‐glucosidase. Furthermore, GSH1 exhibited significant antiobesity effects. In conclusion, GSH1 holds promise as a natural antiobesity ingredient with potential applications in food and pharmaceutical development.
