The effect of perchlorate, thiocyanate and other thyroid-blocking anions upon thyroid hormone-plasma protein interaction of the rat was studied in vitro by 2 methods: the uptake of labeled thyroxine (131I-T4) and triiodothyronine (131I-T3) by muscle and electrophoresis of plasma containing 131I-T4. Increases in the anion concentration and the uptake by muscle were parallel in the presence of dilute plasma, while, in the absence of plasma, there was no anion effect upon muscle uptake. The increaseincrease in muscle uptake paralleled the known activity of each thyroid blocking anion (C1O4-> SCN->BF4>NO3>C1O3). Other anions (Cl-, BrO3- and MnO4-), known to be ineffective in blocking thyroid iodide transport, failed to increase the muscle uptake of 131I-T4 in the presence of plasma. Electrophoresis on acrylamide and starch gel indicated an in vitro displacement of 131I-T4 from prealbumin-albumin area in the presence of thyroid-blocking anions. {Endocrinology82: 47, 1968)
