A method for the radioimmunoassay of arginine vasotocin has been developed using methods and antisera previously utilized in the measurement of serum arginine vasopressin. For ssay of biological samples containing both vasotocin and vasopressin, simultaneous measurement of duplicate samples is conducted with two antisera.Samples known to contain only vasotocin require the use of only one antiserum. In mammalian samples 125I-vasopressin is used as labeled antigen, but in vasopressin-free samples (nonmammalian ertebrate samples) 125I-vasotocin serves as labeled antigen and provides greater sensitivity and specificity. Measurements of pituitary content of vasotocin in chickens and vasopressin in rats provided results consistent with previously reported data. Application to bullfrogs revealed increasing levels of serum immunoreactive vasotocin with increasing dehydration.(Endocrinology95: 1726, 1974)
