Polyribosomes from etiolated soybean (Glycine max L.) hypocotyl tissue were largely separated from ribosomes by collection in the microsomal fraction. Relative polyribosome content was used as an indication of messenger RNA level as affected by 2,4-D. Treated tissue went through a growth-promotive phase (characterized by high messenger RNA synthesis) before an inhibitory phase was reached. The onset of DNA synthesis, whether in stimulated basal tissue or apical tissue after release from inhibition, resulted in a concomitant burst of ribosome production. These results were confirmed by P32 labeling of nucleic acids.
