A previously described method for preparing rat adrenal cell suspensions by use of collagenase has been modified. The sensitivity of the preparation to stimulation by ACTH and 3′,5′-cyclic AMP was investigated. The dibutyryl derivative of 3′,5′-cyclic AMP was 30 times more effective, at the concentrations tested, than the parent compound. Concerning nucleoside/nucleotide specificity of the adrenal cell suspensions, of 42 purine and pyrimidine compounds tested, 3′,5′-cyclic IMP was active, at about 50% of the activity of 3′,5′-cyclic AMP. All of the 3′,5′- cyclic nucleotides tested showed synergism in the presence of 5.0 mM theophylline; the most striking effect was seen with 3′,5′-cyclic GMP. 2′,3′- Cyclic GMP and 2′,3′-cyclic UMP were potent inhibitors of ACTH and 3′,5′-cyclic AMP-stimulated steroidogenesis in preparations of adrenal cell suspensions.
