Approximately 10% of the single-copy sequences of rat-liver chromatin can be excised by DNase-II digestion followed by precipitation with 2 mm MgCl2 (Marushige and Bonner 1971; Billing and Bonner 1972; Gottesfeld et al. 1974b, 1976). We have found by several criteria that the fraction not precipitated is the template-active portion of the genome. First, it contains the bulk of nascent RNA (Billing and Bonner 1972). Second, the DNA of this fraction hybridizes extensively to whole-cell RNA, whereas the remaining 90% does not (Gottesfeld et al. 1974b). Finally, the DNA of the soluble fraction constitutes a 10% subset of the single-copy sequences and a 15–20% subset of the families of repetitive sequences of whole-rat DNA.
