Incorporation of exogenous, radiolabelled alanine, phenylalanine and histidine into the protein fraction of Candida albicans was measured with and without the presence of 5-fluorocytosine (5-FC), in the culture medium. Incorporation of phenylalanine was either increased or decreased with the drug depending on the experimental procedure, but that of alanine was consistently increased and that of histidine, decreased. Interpretation was difficult since an increase in the total pool size and profound changes in the concentration of many endogenous amino acids, including alanine and phenylalanine, in the pool of disrupted fungus cells also took place. However, the concentration of endogenous histidine remained constant, and in addition, the transport of the exogenous amino acid into the cells was not affected. Therefore, the consistent decrease in the incorporation of radiolabelled histidine probably indicates a real influence of 5-FC on protein synthesis in susceptible fungi. This effect may be responsible for, or contribute to, the antifungal activity of the drug.
