Arrdc2 is identified as a novel mechanosensitive gene that promotes myotube atrophy, providing a potential mechanism for disuse muscle atrophy in aging.
Arrdc2 may mediate disuse atrophy; leaves open its role and therapeutic targeting in human aging muscle.
We establish Arrdc2 as a novel mechanosensitive gene highly induced in response to mechanical unloading, particularly in aged muscle. Arrdc2 induction in C2C12 myotubes is sufficient to produce thinner myotubes and a transcriptional landscape consistent with muscle atrophy and disuse.
