A meta-analysis of 15 randomized controlled trials involving 87,549 participants evaluated the impact of GLP-1RAs on cardiovascular outcomes across varying cardiovascular-kidney-metabolic comorbidity.
Why the study?

Dear Editors, The recent study by Siddiqi, T. J. et al. (2025) titled ‘Effect of Glucagon-Like Peptide-1 Receptor Agonists on Heart Failure Outcomes and Cardiovascular Death Across Varying Cardiovascular-Kidney-Metabolic Comorbidity’ offers a comprehensive meta-analysis on the impact of glucagon-like peptide-1 receptor agonists (GLP-1RAs) on cardiovascular outcomes in patients with comorbid heart failure, chronic kidney disease, and metabolic disorders.1 This work is significant for its large sample size and its attempt to quantify the potential benefits of GLP-1RAs in a clinically relevant patient cohort. The authors performed an extensive review of 15 randomized controlled trials involving a total of 87 549 participants. The study’s strength lies in its rigorous statistical approach, using random-effects models to analyse the hazard ratios (HR) and relative risks across different subgroups. This is particularly important for a drug class like GLP-1RAs, which has widespread implications in the treatment of type 2 diabetes (T2DM), obesity, and heart failure.
