The problem of uneven hematoxylin and eosin (H&E) staining is a widespread phenomenon. When water due to incomplete clearing, as in a humid environment, was found to be the problem, substitution of toluene for xylene in the tissue processing procedure eliminated the artifact. The importance of an effective quality assurance in identifying equipment malfunction or contamination of reagents as a cause of uneven staining is stressed. (The J Histotechnol 13:193, 1990)
