Responses of runaway replication to various rates of temperature change were studied both experimentally and theoretically. When the temperature shift was completed within 30 minutes, maximum plasmid replication rate and production of gene products were observed as in the stepwise temperature change. Runaway replication could be induced fairly well, as long as the temperature‐shift was completed within 2 hours. A kinetic model of the runaway replication was proposed and it expressed the characteristics of a recombinant containing a plasmid very well. These results indicate that a runaway‐replication plasmid can be applied to industrial production of gene products.
