The separation of two types of catalytic activity exhibited by purine nucleoside phosphorylase purified from Ehrlich ascites tumor cells is described. Fractions catalyzing either phosphorolysis or ribosyl transfer were obtained by sucrose gradient centrifugation. Data regarding the conversion of phosphorolytic to transfer activity are presented, and observations relevant to the expression of the two catalytic activities are briefly discussed.
