An accumulation of xanthine and a decrease of uric acid formation as the effect of 4‐hydroxy‐pyrazolo[3,4‐d]pyrimidine (allopurinol) and other inhibitors (8‐azaadenine, 8‐azaguanine, 6‐thio‐purine) on hypoxanthine hydroxylation by milk xanthine oxidase in vitro was found. A remaining activity of xanthine oxidoreductases seems to be the main base of enhanced formation of xanthine due to allopurinol treatment in vivo. Xanthine oxidoreductases, increasing and decreasing the pool of oxypurines (hypoxanthine and xanthine) must play an important role in purine biosynthesis regulation.
